RESUMO

The presence of nanoplastics (NPs) and microplastics (MPs) in the environment is recognised as a global-scale problem. Due to their hydrophobic nature and large specific surface, NPs and MPs can adsorb other contaminants, as polycyclic aromatic hydrocarbons (PAHs), and modulate their bioavailability and hazard. Adult zebrafish were exposed for 3 and 21 days to: (1) 0.07 mg/L NPs (50 nm), (2) 0.05 mg/L MPs (4.5 µm), (3) MPs with sorbed oil compounds of the water accommodated fraction (WAF) of a naphthenic crude oil (MPs-WAF), (4) MPs with sorbed benzo(a)pyrene (MPs-B(a)P), (5) 5 % WAF and (6) 21 µg/L B(a)P. Electrodense particles resembling NPs were seen in the intestine lumen close to microvilli. MPs were abundantly found in the intestine lumen, but not internalised into the tissues. After 21 days, NPs caused a significant downregulation of cat, and upregulation of gpx1a and sod1, while MPs upregulated cyp1a and increased the prevalence of liver vacuolisation. No histopathological alteration was observed in gills. In this study, contaminated MPs did not increase PAH levels in zebrafish but results highlight the potential differential impact of plastic particles depending on their size, making it necessary to urgently address the ecotoxicological impact of real environmental NPs and MPs.

In vertebrates, fasting is an intricate physiological process associated with strong metabolic changes, yet its effect on pollutant residue variation is poorly understood. Here, we quantified long-term changes in plasma concentrations of 20 organochlorine and 16 perfluoroalkyl pollutants in king penguins Aptenodytes patagonicus during the breeding and molting fasts, which are marked by low and high levels of protein catabolism, respectively, and by strong lipid use. The profile of measured pollutants in plasma was dominated by perfluorooctanesulfonic acid (PFOS, initial relative contribution of 60%). Initial total pollutant concentrations were similar in molting (3.3-5.7 ng g-1 ww) and breeding penguins (range of 4.2-7.3 ng g-1 wet weight, ww). Long-term fasting (25 days) for molting and breeding led, respectively, to a 1.8- and 2.2-fold increase in total plasma pollutant concentrations, although the rate and direction of change were compound-specific. Hexachlorbenzene (HCB) and PFOS concentrations increased in plasma (net mobilization) during both types of fasting, likely due to lipid use. Plasma perfluoroundecanoate (PFUnDA) and perfluorotridecanoate (PFTrDA) concentrations increased in breeders (net mobilization) but decreased in molting individuals (net excretion), suggesting a significant incorporation of these pollutants into feathers. This study is a key contribution to our understanding of pollutant variation in blood during long-term fasting in wildlife.

On September 26th 2019, a major fire occurred in the Lubrizol factory located near the Seine estuary, in Rouen-France. Juvenile flounders were captured in the Canche estuary (a reference system) and caged one month in the Canche and in the Seine downstream the accident site. No significant increases of PAHs, PCBs and PFAS was detected in Seine vs Canche sediments after the accident, but a significant increase of dioxins and furans was observed in water and sewage sludge in the Rouen wastewater treatment plant. The proteomics approach highlighted a dysregulation of proteins associated with cholesterol synthesis and lipid metabolism, in fish caged in the Seine. The overall results suggested that the fire produced air borne dioxins and furans that got deposited on soil and subsequently entered in the Seine estuarine waters via runoff; thus contaminating fish preys and caged flounders in the Seine estuary.

Akanda National Park (ANP) is composed of mangrove ecosystems bordering Libreville, Gabon's capital. The contamination of aquatic resources from the ANP by persistent organic pollutants (POPs) and trace metals (TMs) was never evaluated. To provide a basis for their monitoring in the ANP, five species (two fish, two mollusks, and one crustacean) were analyzed from three sampling sites in 2017. Contamination levels for POPs and TMs were below maximum acceptable limits for seafood, including Cd and Pb. No DDT was found in any sample. Inter-specific differences were more obvious than the differences among sites, although the results may be biased by an unbalanced sampling design. The oyster Crassostrea gasar was the most contaminated species, making this species a good candidate to assess environmental contamination in the area. The studied species also contained essential elements, such as Fe, Zn and Mn at interesting levels in a nutritional point of view.

A collaborative trial involving 16 participants from nine European countries was conducted within the NORMAN network in efforts to harmonise suspect and non-target screening of environmental contaminants in whole fish samples of bream (Abramis brama). Participants were provided with freeze-dried, homogenised fish samples from a contaminated and a reference site, extracts (spiked and non-spiked) and reference sample preparation protocols for liquid chromatography (LC) and gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS). Participants extracted fish samples using their in-house sample preparation method and/or the protocol provided. Participants correctly identified 9-69 % of spiked compounds using LC-HRMS and 20-60 % of spiked compounds using GC-HRMS. From the contaminated site, suspect screening with participants' own suspect lists led to putative identification of on average ∼145 and ∼20 unique features per participant using LC-HRMS and GC-HRMS, respectively, while non-target screening identified on average ∼42 and ∼56 unique features per participant using LC-HRMS and GC-HRMS, respectively. Within the same sub-group of sample preparation method, only a few features were identified by at least two participants in suspect screening (16 features using LC-HRMS, 0 features using GC-HRMS) and non-target screening (0 features using LC-HRMS, 2 features using GC-HRMS). The compounds identified had log octanol/water partition coefficient (KOW) values from -9.9 to 16 and mass-to-charge ratios (m/z) of 68 to 761 (LC-HRMS and GC-HRMS). A significant linear trend was found between log KOW and m/z for the GC-HRMS data. Overall, these findings indicate that differences in screening results are mainly due to the data analysis workflows used by different participants. Further work is needed to harmonise the results obtained when applying suspect and non-target screening approaches to environmental biota samples.

Ultrasound as a green and efficient process gains special attention in wastewater treatment. The ultrasound-assisted degradation of azoxystrobin, methoxyfenozide, and propyzamide as widely used pesticides for vine treatment was investigated. A wide range of ultrasonic power (40 to 140 W) and a single frequency (20 kHz) were applied. Degradation experiments were carried out according to the parameters set by a central composite design (CCD) under response surface methodology (RSM) via JMP software. The treatment efficiency was quantified using degradation rates and hydrogen peroxide (H2O2) measurements. Results indicated that the pesticide's degradation was negligible at 40 W but by increasing the power setting from 80 to 140 W, the degradation rate constants of azoxystrobin, methoxyfenozide, and propyzamide increased from 3.6 × 10-2 min-1 to 0.2 min-1, from 6.1 × 10-2 min-1 to 0.3 min-1, and from 3.1 × 10-2 min-1 to 0.1 min-1, respectively. The hydrogen peroxide (H2O2) measurements confirmed this trend. Besides, electric energy per order of pollutant removal (EE/O) was also evaluated for the same treatment duration and results revealed that treatment conditions of 20 kHz and 140 W were the less energy-guzzling. Finally, profiles obtained with RSM illustrated linear degradation kinetics for azoxystrobin and propyzamide. Indeed, treatment efficiency increased when increasing both studied parameters. However, both linear and quadratic degradation kinetics occurred for methoxyfenozide degradation indicating a parameter threshold beyond which the trend is reversed. Overall, this study confirms the effectiveness of ultrasound for the degradation of pesticides in aqueous medium.

Aquatic environments are the final receptors of human emissions and are therefore contaminated by molecules, such as pharmaceuticals. After use, these compounds and their metabolites are discharged to wastewater treatment plants (WWTPs). During wastewater treatment, compounds may be eliminated or degraded into transformation products (TPs) or may be persistent. The aim of this study was to develop an analytical method based on high resolution mass spectrometry (HRMS) for the identification of six psychotropic drugs that are widely consumed in France and present in WWTPs, as well as their potential associated metabolites and TPs. Four out of six psychotropic drugs and between twenty-five and thirty-seven potential TPs were detected in wastewater, although this was based on full scan data. TPs not reported in the literature and specific to the study sites and therefore to the wastewater treatment processes were tentatively identified. For the selected drugs, most known and present TPs were identified, such as desmethylvenlafaxine or norcitalopram. Moreover, the short fragmentation study led rather to the identification of several TPs of carbamazepine as ubiquitous persistent TPs.

Triazoles belong to a family of fungicides that are ubiquitous in agroecosystems due to their widespread use in crops. Despite their efficiency in controlling fungal diseases, triazoles are also suspected to affect non-target vertebrate species through the disruption of key physiological mechanisms. Most studies so far have focused on aquatic animal models, and the potential impact of triazoles on terrestrial vertebrates has been overlooked despite their relevance as sentinel species of contaminated agroecosystems. Here, we examined the impact of tebuconazole on the thyroid endocrine axis, associated phenotypic traits (plumage quality and body condition) and sperm quality in wild-caught house sparrows (Passer domesticus). We experimentally exposed house sparrows to realistic concentrations of tebuconazole under controlled conditions and tested the impact of this exposure on the levels of thyroid hormones (T3 and T4), feather quality (size and density), body condition and sperm morphology. We found that exposure to tebuconazole caused a significant decrease in T4 levels, suggesting that this azole affects the thyroid endocrine axis, although T3 levels did not differ between control and exposed sparrows. Importantly, we also found that exposed females had an altered plumage structure (larger but less dense feathers) relative to control females. The impact of tebuconazole on body condition was dependent on the duration of exposure and the sex of individuals. Finally, we did not show any effect of exposure to tebuconazole on sperm morphology. Our study demonstrates for the first time that exposure to tebuconazole can alter the thyroid axis of wild birds, impact their plumage quality and potentially affect their body condition. Further endocrine and transcriptomic studies are now needed not only to understand the underlying mechanistic effects of tebuconazole on these variables, but also to further investigate their ultimate consequences on performance (i.e. reproduction and survival).

Contamination of the environment by pesticide residues is a growing concern given their widespread presence in the environment and their effects on ecosystems. Only a few studies have addressed the occurrence of pesticides in soils, and their results highlighted the need for further research on the persistence and risks induced by those substances. We monitored 111 pesticide residues (48 fungicides, 36 herbicides, 25 insecticides and/or acaricides, and two safeners) in 47 soils sampled across France under various land uses (arable lands, vineyards, orchards, forests, grasslands, and brownfields). Pesticides were found in 98% of the sites (46 of the 47 sampled), including untreated areas such as organic fields, forests, grasslands, and brownfields, with up to 33 different substances detected in one sample, mostly fungicides and herbicides. The concentrations of herbicides were the highest in soils with glyphosate, and its transformation product, AMPA, contributed 70% of the cumulative herbicides. Risk assessment underlined a moderate to high risk for earthworms in arable soils mostly attributed to insecticides and/or acaricides. Finally, the comparison with pesticide application by farmers underlines the presence of some residues long after their supposed 90% degradation and at concentrations higher than predicted environmental concentrations, leading to questions their real persistence in soils.

In this paper, we investigate for the first time the contamination source and the ecological risk associated to organochlorinated compounds in the Litani system. For this purpose, the levels of 7 polychlorinated biphenyls (PCBs) and 13 organochlorinated pesticides (OCPs) were assessed, using a microwave-assisted extraction coupled to gas chromatography-electron capture detector (MAE/GC-ECD) method, in surficial sediments from 30 sites along the main course of the river, two major tributaries, and the Quaraoun Lake. ∑7PCBs exhibited total concentrations ranging from 0.11 to 8 ng g-1 of dry weight and are not able apparently to pose ecological risks since none of the samples showed concentration above the effects range low (ERL) guideline (22.7 ng g-1). The detected levels of OCPs in the river were significantly higher than those of PCBs; ∑13OCPs range from 0.5 to 46.5 ng g-1 of dry weight. Overall, the integrated eco-toxicological risk imposed by the organochlorine contamination in the Litani River, estimated as the mean effects range medium quotient (mERMq), is considered low with risk probability lower than 21% in all sites. The six dichlorodiphenyltrichloroethane-based pesticides (∑6DDT) contributed to more than 70% of the mERMq in 15 over the 29 included sites. Moreover, the approach of ∑6DDT/∑7PCB ratios was applied to investigate the contamination source. ∑6DDT/∑7PCB was higher than one in all samples suggesting that the organochlorine contamination result from agricultural activities set in the surrounding areas of the Litani River.

The PESTIPREV study has been designed to investigate residential exposure to pesticides applied to vines and ultimately propose mitigation measures. A feasibility study was carried out to validate a protocol for measuring six pesticides in three houses located near vineyards in July 2020. Samples included indoor and outdoor surfaces sampled with wipes (n = 214), patches on the resident's skin (n = 7), hand or foot washing (n = 5), and pets sampled using wipes (n = 2). Limits of quantification for wipes ranged between 0.02 ng for trifloxystrobin and 1.50 ng for pyraclostrobin. Tebuconazole and trifloxystrobin were quantified in nearly 100% of the surface samples, whereas the other fungicides were less frequently found (from 39.7% for pyraclostrobin to 55.1% for boscalid). The median surface loadings ranged from 3.13 ng/m2 for benalaxyl to 82.48 ng/m2 for cymoxanil. The pesticides most frequently quantified in hand washing, patch samples, and pet wipes were the same as those quantified on surfaces. Finally, the analyses proved to be successful. The tools developed to collect information on determinants were well completed. The protocol was well received by the participants and appeared to be feasible and relevant to the objective of the PESTIPREV study, although some improvements have been identified. It was applied on a larger scale in 2021 to study the determinants of pesticide exposure.

Human-induced climate change impacts the oceans, increasing their temperature, changing their circulation and chemical properties, and affecting marine ecosystems. Like most marine species, sole has a biphasic life cycle, where one planktonic larval stage and juvenile/adult stages occur in a different ecological niche. The year-class strength, usually quantified by the end of the larvae stage, is crucial for explaining the species' recruitment. We implemented an experimental system for rearing larvae under laboratory conditions and experimentally investigated the effects of temperature and feeding frequencies on survival, development (growth), and metamorphosis success of S. solea larvae. Specific questions addressed in this work include: what are the effects of feeding regimes on larvae development? How does temperature impact larvae development? Our results highlight that survival depends on the first feeding, that the onset of metamorphosis varies according to rearing temperature and that poorly fed larvae take significantly longer to start (if they do) metamorphosing. Moreover, larvae reared at the higher temperature (a +4°C scenario) showed a higher incidence in metamorphosis defects. We discuss the implications of our results in an ecological context, notably in terms of recruitment and settlement. Understanding the processes that regulate the abundance of wild populations is of primary importance, especially if these populations are living resources exploited by humans.

While perfluoroalkyl sulfonic acids (PFSAs) and perfluoroalkyl carboxylic acids (PFCAs) are ubiquitous in aquatic environments, non-targeted methods have recently revealed the presence of numerous unidentified per- and polyfluoroalkyl substances (PFAS). Besides those methods, the total oxidizable precursor (TOP) assay has proved useful to estimate the contribution of unattributed perfluoroalkyl acids precursors (pre-PFAAs). In this study, an optimized extraction method was developed to examine the spatial distribution of 36 targeted PFAS in surface sediments collected at French nationwide scale (n = 43), including neutral, anionic and zwitterionic molecules. In addition, a TOP assay procedure was implemented to estimate the contribution of unattributed pre-PFAAs in these samples. Conversion yields of targeted pre-PFAAs were determined for the first time under realistic conditions and led to differences in oxidation profiles compared to the common spiked ultra-pure water method. PFAS were detected in 86 % of samples and ∑PFAStargeted was in the range < Limit of Detection - 23 ng g-1 dry weight (dw) (median: 1.3 ng g-1 dw), with ∑pre-PFAAstargeted representing on average 29 ± 26 % of ∑PFAS. Among pre-PFAAs, compounds of emerging interest such as the fluorotelomer sulfonamidoalkyl betaines 6:2 FTAB and 8:2 FTAB were respectively detected in 38 % and 24 % of samples, with levels similar to those of L-PFOS (<0.36-2.2, <0.50-6.8 and < 0.08-5.1 ng g-1 dw, respectively). A hierarchical cluster analysis coupled with a geographic information system-based approach revealed similarities between groups of sampling sites. For instance, elevated contribution of FTABs were associated with the proximity to airport activities where betaine-based aqueous film-forming foam (AFFFs) might have been used. In addition, unattributed pre-PFAAs were strongly correlated with ∑PFAStargeted and they accounted for 58 % of ∑PFAS (median value); they were generally found in larger quantity near industrial and urban areas where the highest ∑PFAStargeted were also observed.

Polychlorinated biphenyls (PCBs) are recognized as persistent organic pollutants and accumulate in organisms, soils, waters, and sediments, causing major health and ecological perturbations. Literature reported PCB bio-transformation by fungi and bacteria in vitro, but data about the in situ impact of those compounds on microbial communities remained scarce while being useful to guide biotransformation assays. The present work investigated for the first time microbial diversity from the three-domains-of-life in a long-term contaminated brownfield (a former factory land). Soil samples were ranked according to their PCB concentrations, and a significant increase in abundance was shown according to increased concentrations. Microbial communities structure showed a segregation from the least to the most PCB-polluted samples. Among the identified microorganisms, Bacteria belonging to Gammaproteobacteria class, as well as Fungi affiliated to Saccharomycetes class or Pleurotaceae family, including some species known to transform some PCBs were abundantly retrieved in the highly polluted soil samples.

To assess the risk of pesticide mixtures in lagoon waters, this study adopted a multi-step approach using integrative passive samplers (POCIS) and concentration addition (CA) toxicological models. Two French Mediterranean lagoons (Thau and Or) were monitored for a range of 68 pesticides continuously over a period of a year (2015-16). The findings revealed mixtures of dissolved pesticides with varying composition and levels over the year. The Or site contained more pesticides than Thau site (37 vs 28 different substances), at higher concentrations (0.1-58.6 ng.L-1 at Or vs <0.1-9.9 at Thau) and with overall higher detection frequencies. All samples showed a potential chronic toxicity risk, depending on the composition and concentrations of co-occurring pesticides. In 74 % of the samples, this pesticide risk was driven by a few single substances (ametryn, atrazine, azoxystrobin, carbendazim, chlorotoluron, irgarol, diuron and metolachlor) and certain transformation products (e.g. DPMU and metolachlor OA/ESA). Individually, these were a threat for the three taxa studied (phytoplankton, crustaceans and fish). Yet even a drastic reduction of these drivers alone (up to 5 % of their current concentration) would not eliminate the toxicity risks in 56 % of the Or Lagoon samples, due to pesticide mixtures. The two CA-based approaches used to assess the combined effect of these mixtures, determined chronic potential negative impacts for both lagoons, while no acute risk was highlighted. This risk was seasonal, indicating the importance of monitoring in key periods (summer, winter and spring) to get a more realistic picture of the pesticide threat in lagoon waters. These findings suggest that it is crucial to review the current EU Water Framework Directive's risk-assessment method, as it may incorrectly determine pesticide risk in lagoons.

Azoles represent the most used family of organic fungicides worldwide and they are used in agriculture to circumvent the detrimental impact of fungi on yields. Although it is known that these triazoles can contaminate the air, the soil, and the water, field data are currently and dramatically lacking to assess if, and to what extent, the use of triazoles could contaminate non-target wild vertebrate species, notably in agroecosystems. In this study, we aimed to document for the first time the degree of blood contamination of a generalist wild bird species by multiple azoles which are used for plant protection and fungi pest control in various habitats. We deployed passive air samplers and captured 118 Common blackbirds (Turdus merula) in an agroecosystem (vineyard), a protected forest, and a city in western France. We collected blood and analyzed the plasma levels of 13 triazoles and 2 imidazoles. We found that a significant percentage of blackbirds living in vineyards have extremely high plasma levels of multiple azoles (means (pg.g-1); tebuconazole: 149.23, difenoconazole: 44.27, fenbuconazole: 239.38, tetraconazole: 1194.16), while contamination was very limited in the blackbirds from the protected forest and absent in urban blackbirds. Interestingly, we also report that the contamination of blackbirds living in vineyard was especially high at the end of Spring and the beginning of Summer and this matches perfectly with the results from the passive air samplers (i.e., high levels of azoles in the air of vineyards during June and July). However, we did not find any correlation between the levels of plasma contamination by azoles and two simple integrative biomarkers of health (feather density and body condition) in this sentinel species. Future experimental studies are now needed to assess the potential sub-lethal effects of such levels of contamination on the physiology of non-target vertebrate species.

In fish, the gut microbiome plays a crucial role in homeostasis and health and is affected by several organic and inorganic environmental contaminants. Amphidromous fish are sentinel species, particularly exposed to these stressors. We used whole metagenome sequencing to characterize the gut microbiome of wild European eels (Anguilla anguilla) at a juvenile stage captured from three sites with contrasted pollution levels in term of heavy metals and persistent organic pollutants. The objectives were to identify what parameters could alter the gut microbiome of this catadromous fish and to explore the potential use of microbiota as bioindicators of environment quality. We identified a total of 1079 microbial genera. Overall, gut microbiome was dominated by Proteobacteria, Firmicutes and Actinobacteria. Alpha and beta diversity were different amongst sites and could be explained by a reduced number of environmental and biological factors, specifically the relative abundance of fish preys in eels' diet, PCB101, γHCH (lindane), transnonachlor and arsenic. Furthermore, we identified a series of indicator taxa with differential abundance between the three sites. Changes in the microbial communities in the gut caused by environmental pollutants were previously undocumented in European eels. Our results indicate that microbiota might represent another route by which pollutants affect the health of these aquatic sentinel organisms.

Triazole compounds are among the most widely used fungicides in agroecosystems to protect crops from potential fungal diseases. Triazoles are suspected to have an impact on nontarget species due to their interactions with nonfungal sterol synthesis, and wild birds are likely to be contaminated by triazole fungicides because many of them live in agroecosystems. We experimentally tested whether exposure to environmental concentrations of a triazole could alter key integrative traits (metabolic rates and body condition) of an agroecosystem sentinel species, the house sparrow (Passer domesticus). Wild-caught adult sparrows were maintained in captivity and exposed (exposed group) or not (control group) for 7 continuous months to tebuconazole through drinking water. The metabolic rates of exposed and control sparrows were then measured at two different temperatures (12 °C and 25 °C), which correspond, respectively, to the thermoregulation and thermoneutrality temperatures of this species. We found that exposed sparrows had lower resting metabolic rates (i.e., measured at thermoneutrality, 25 °C) than controls. However, the thermoregulatory metabolic rates (i.e., measured at 12 °C) did not differ between exposed and control sparrows. Although the body mass and condition were not measured at the beginning of the exposure, sparrows at the time of the metabolic measurements 7 months after the onset of such exposure had a higher body condition than controls, supporting further the idea that tebuconazole affects metabolic functions. Our study demonstrates for the first time that the use of tebuconazole can alter metabolism and could potentially lead to adverse effects in birds. The present study, conducted in the Galion Bay in Martinique, aims to highlight the temporal and seasonal variations of chlordecone contamination (an organochlorine pollutant) in the ambient environment (seawater) and also in the marine organisms in three main coastal marine habitats (mangroves, seagrass beds and coral reefs). To this end, two methodologies were used to measure and compare the chemical contamination of seawater during 13 months (spot samplings and POCIS technique). In parallel, concentrations of chlordecone and isotopic ratios (C and N) were carried out on marine organisms, collected during two contrasting climatic periods (dry and rainy), to evidence seasonal variations. The results showed that the contamination of seawater displayed significant variations over time and depended on environmental factors such as water flows, which imply dilution and dispersion phenomena. Concerning the marine organisms, the level of contamination varied considerably between the two seasons in seagrass beds with higher levels of contamination during the rainy season. Reef organisms were more moderately affected by this pollution, while mangrove organisms showed a high level of chlordecone whatever the season. Finally, isotope analyses highlighted that bioamplification along marine food webs occurs at each season and each station.
